Effect of controlled released PTH (1-34) encapsulated PLGA microspheres on treatment of early osteoarthritis in rat

چکیده

INTRODUCTION: Parathyroid hormone (PTH) is an 84 amino acid polypeptide that acts as the most important regulator of calcium homeostasis in the human body through its direct action on bone and kidney. Previously we reported that PTH(1-34) inhibits terminal differentiation of articular chondrocytes and in turn suppresses the progression of osteoarthritis (OA). However, the treatment needs the injection of PTH every 3 days over the treatment period. We hypothesized that encapsulation of PTH(1-34) in PLGA microspheres with controlled release property may prolong the injection durations and can be used to treat OA with less injection frequency. Here in we studied the effect of released PTH(1-34) from PLGA(65:35) encapsulated PTH(1-34) (PLGA/PTH) microspheres on the papain-induced OA in the rat knee joints. In vitro study, we studied the microspheres morphology, PTH(1-34) encapsulation efficiency, release profile, bioactivity. In vivo study, we evaluated the changes of localized glycosaminoglycan (GAG), Type II collagen and Type X collagen in the articular cartilage of rat knee.

Preparation and In-vitro Evaluation of Controlled Release PLGA Microparticles Containing Triptoreline

Triptoreline is a potent agonist of luteinizing hormone-releasing hormone, currently used in the treatment of prostatic cancer where therapy may be required over months or years. Frequent injection of drug decreases patients’ compliance. The present study describes the formulation of a sustained release microparticulate drug delivery system containing triptoreline acetate, using poly (D,L lacti...

Incorporation of protein-eluting microspheres into biodegradable nerve guidance channels for controlled release.

Nerve guidance channels (NGCs) promote axonal regeneration after transection injury of the peripheral nerve or spinal cord, yet this regeneration is limited. To enhance regeneration further, we hypothesize that localized delivery of therapeutic molecules combined with the NGC is required. In an attempt to achieve such an NGC, we designed and synthesized a novel NGC in which protein-encapsulated...

Sustained Release of Risedronate from PLGA Microparticles Embedded in Alginate Hydrogel for Treatment of Bony Lesions

Background: Inflammatory bone resorption in periodontitis can lead to tooth loss. Systemic administration of bisphosphonates such as risedronate for preventing bone resorption can cause adverse effects. Alginate hydrogel (ALG) and poly (lactic acid-co-glycolic acid) (PLGA) microparticles have been studied as drug delivery systems for sustained release of drugs. Therefore, the release pattern of...

In vitro and in vivo evaluation of a somatostatin analogue released from PLGA microspheres.

The purpose of this study was to design poly(lactide-co-glycolide) (PLGA) microspheres for the continuous delivery of the somatostatin analogue, vapreotide, over 2-4 weeks. The microspheres were produced by spray-drying and the desired characteristics, i.e. high encapsulation efficiency and controlled release over 2-4 weeks, achieved through optimizing the type of polymer, processing solvent, a...
